Spooling braid with mono

Can I tie any knot via braid on mono before spooling the braid? Sorry if this is confusing. (Never used braided line and I’m currently in the process of putting it on. 20lb) Update: Got the knots tied on and spooled. It casts great.

A Double uni knot is dependable for this and for leader lines.
